According to local government officials, the incident took place around 9:30 pm local time leaving the district police chief Abdul Manaf dead along with two policemen.
Provincial police spokesman, Zia Durani said at least six policemen were also injured following the suicide attack.
Durani further added that the injured policemen were taken to hospital and their health condition have reported as satisfactory.
He said the suicide bomber detonated his explosives after he managed to enter the district police department.
No group including the Taliban militants has so far claimed responsibility behind the incident.
Kandahar is among the volatile provinces in southern Afghanistan where anti-government armed militant groups are actively operating and frequently carry out insurgency activities.
The district police chief for Spin Boldak was also killed following an infighting between the policemen earlier last month.
